The history of Jamestown is rich. Originally inhabited by the Narragansett, settled in 1638 by the English, the island was used for grazing sheep. In 1678, Conanicut Island was incorporated as the town of Jamestown. In 1728, the town built a windmill for grinding corn. On December 10, 1775, 200 British and Hessian troops landed at East Ferry on Conanicut Island and marched to West Ferry, where they burned the ferry house. As they returned to East Ferry, they destroyed many buildings, including fourteen homes; which caused more than 200 of Conanicut Island's 556 residents to flee to the mainland. A number of signers of the Declaration of Independence can be viewed on a document in Town Hall.

The Jamestown Historical Society's museum building was originally built in 1885 as a one-room primary school, later serving as the town's library. In 2008 the interior of the building was refurbished. Each year, the Jamestown Historical Society creates a new exhibit for display. The Summer 2018 exhibit is Then & Now - Where We Stayed including old postcards and photographs of the hotels and boarding houses of Jamestown.
